The influence of different concentration and centrifugal parameters on TEM sample preparation of Escherichia coli

Abstract:
Objective:To investigate the influences of different OD value and centrifugal parameters on the ultrastructure of Escherichia coli in order to find out the best OD value,speed and time of centrifugation.Methods:According to orthogonal experimental designing principle,the E.coli with different OD600 values are divided into 9 different groups of centrifuge speed and time.The changes of E.coli morphology in each group at different OD600 value,centrifuge time and speed were compared under transmission electron microscope.Results:The different OD600 values and centrifugal parameters have little influence on the ultrastructure of E.coli,but when the OD600 value at 0.8,centrifuge time at 15 min,the cross-section ratio was significantly higher than other groups and was more suitable for observation.Conclusion:The best parameter to prepare the TEM samples of E.coli is under the OD600 value at 0.8 and centrifugation time at 8 minutes and speed at 5 000～12 000 r/min.
