Abstract:
Objective:To develop a novel amperometic immunosensor for the determination of Urinary Trypsinogen-2(TSG-Ⅱ).Methods:the immunosensor was developed by modifying Nafion film on the glassy carbon electrode(GCE),then thionine and colloidal gold were immobilized by static electricity and covalent bonding self-assembly.And then the antibody of Urinary TSG-Ⅱwas absorbed on the surface of nano-gold monolayer. Finally,Horseradish peroxidase(HRP) blocked the non-specific adsorption site on the electrode.Result:Experiment results showed that the good linear relations between peak current and concentration of TSG-Ⅱ were established in the range of 6.0-100?g/L.The detection limit was 3.5?g/L.Conclusion:The amperometie immunoassay biosensor has high sensitivity and good specificity and it has laid the foundation to establish a new method for detection of TSG-Ⅱ.
