Abstract:
Objective:To investigate the interventional effect of rosiglitazone on the expression of protein ABCA1(ATP-binding cassette transporter-1) in peripheral blood monocytes(PBMC) in patients with acute coronary syndrome(ACS).Methods:Twenty-one patients with ACS were enrolled in the study.PBMCs were isolated by density gradient centrifugation.The experiments included:(1) Rosiglitazone of different final concentrations(0 ?mol/L,1.0 ?mol/L,10?mol/L,100 ?mol/L)were incubated with PBMCs for 24h.(2) Rosiglitazone of the chosen final concentration of 10?mol/L was incubated with PBMCs for 0 h,6 h,12 h and 24 h,separately.(3) Control groups were also set(cells treated with PBS).Three wells served as measured wells.And one well acted as blank well in each group.Then the level of protein ABCA1 was determined by flow cytometry after incubation.Results:Compared with control groups,after PBMCs were intervened by rosiglitazone,the expression of protein ABCA1 of PBMCs increased in dose-and timedepe-ndent manners.Conclusion:Rosiglitazone can increase the expression of protein ABCA1 of PBMCs,which may be one of the possible mechanisms for rosiglitazone to play an anti-atherosclerotic role.
