Abstract:
Objective:To investigate the effect of the silence of Bcl-2 oncogene on the apoptosis of cisplatin-resistant cell line Acc-3/CDDP.Methods:The recombined RNAi plasmids for Bcl-2 oncogene were constituted by the four successive steps-designing of Oligo DNAs,and were transfected into Acc-3/CDDP cells by culturing together. The apoptosis of each cells were evaluated by MTT test, electron microscope, and AO/EB fluorescent dyes respectively. Results:The OD value of RNAi group decreased with time last by MTT test, and dropped to the lowest at 48 h.The apoptosis of RNAi group were observable through electron microscope and fluorescent dyes. The apoptosis rate increased with time from 24 to 72 h after transfection. The apoptosis on each cell cycle were clear and classical. Conclusion:Recombined interfering plasmids can effectively silence the expression of Bcl-2 oncogenes, and the apoptosis of cisplatin-resistant cell line Acc-3/CDDP cells can be enhanced.
