Abstract:
Objective:To develop a method for the determination of vincristini sulfa(sVCR) in tumor cells culture media and intracellular fluid by high performance liquid chromatography(HPLC).Methods:VCR can be separated on C18(4.6 mm?250 mm,5?m)column by using 0.02 mol/L potassium dihydrogen phosphate:methano(l30:70)(pH=6.6)as mobile phase with UV detector at a wavelength of 267 nm.The flow rate was 0.8 ml/min.Results:The standard curve was linear in the concentration range 12.5～160.0?g/m(lr=0.997 4, n=7)and 0.125～12.5?g/m(lr=0.998 5,n=6).The average extracting recovery and average recovery of VCR in tumor cells culture media, RSD of intra-day and inter-day was 73.84%,99.83%,3.14%and 3.99%respectively.The RSD of intra-day and inter-day in intracellular fluid was 2.15%and 1.77%respectively.The detection limit was 0.04?g/ml.Conclusion:This method is simple,rapid,accurate and selective for the determination of VCR.
