Determination of cisatracurium in the plasma of rabbits by HPLC coupled with fluorescence detection

Abstract:
Objective:To determinate the concentration of Cisatracurium in the plasma of an rabbit via a proposed HPLC method. Methods:A C18 column was used,the mobile phase was made up of 50mmol/L dipotassium hydrogen phosphate(pH=3.0) and acetonitrile(50∶50),and examination and emission wavelengths are 200nm and 320nm,respectively. Results:the linear range was 25~2 000ng/ml,the limit of detection(LOD) is 25ng/ml, the intra- and inter-precisions(RSD) were lower than2.56% and 3.68%,respectively. Besides,the recovery of extraction was 85.2%~86.1%(n=5) and the recovery of addition was 93.4%~98.8%. Conclusion:The proposed method proved to be accurate and repetitive.
