The responses of microvascular endothelial cells from human ovary carcinoma to PDGF-BB

Abstract:
Objective:To investigate the difference of biological characteristics and responses to PDGF-BB between human ovarian carcinoma-derived microvascular endothelial cells and human umbilical vein endothelial cells.Methods:The ovarian carcinoma-derived microvascular endothelial cells were isolated and purifed from ovarian carcinoma tissues.Compared to the human umbilical vein endothelial cells,the biological characteristics and responses to PDGF-BB were observed in OdMEC for the changes of growth,PCNA and VEGF expression.Results:20ng/ml PDGF-BB could enhance both kinds of endothelial cells to proliferate,express PCNA and VEGF.The proliferation and PCNA protein expression in HUVEC were augmented more significantly than those in OdMEC,the protein and mRNA of VEGF expression were augmented more significantly than those in HUVEC.Conclusion:The responses of OdMEC to PDGF-BB were significantly different from those of HUVEC,which provides experiment basis for research on ovarian carcinoma vascularization and antiangiogenesis therapy.
