Abstract:
Objective To investigate the spatiotemporal patterns and age-related changes of expression of type Ⅰ and type Ⅳ collagen in the corneal development of Bufo raddei larvae during metamorphosis.Methods Using immunohistochemical method,type Ⅰ and Ⅳ collagen distributions were investigated in the cornea of Bufo raddei at hind-limb bud stage,hind-limb developing stage,forelimb bud stage,complete metamorphosis stage,one-year-old adult and three-year-old adult,respectively.Results The inner and outer cornea fused together into mature cornea during metamorphosis.Type Ⅰ collagen was expressed widely during the period,it existed in Bowman's membrane,Descemet's membrane,corneal stroma and endothelium,especially showing a progressive change in corneal stroma.The expression of type Ⅳ collagen was only observed in Bowman's membrane and Descemet's membrane at first,since the forelimb bud stage,it also expressed in corneal stroma and endothelium,but it didn't express the same as Type Ⅰ.Conclusion Type Ⅰ collagen has the role in the construction of the corneal stroma and its fuction in the corneal development of Bufo raddei larvae during metamorphosis.Type Ⅳ and type Ⅰ collagen expressed differently in corneal stroma showing that the construction of the corneal stroma may depend partly on the right expression proportion of different collagen type.
