Abstract:
Objective:To investigate the effects of oxaliplatin on human leukemia K562 cell and its mechanism.Methods:The leukemia cell growth was measured by MTT-microculture tetrazolium assay.Cell-cycle kinetics and apoptosis were analyzed by flow cytometry and microscopic examination.Results:Oxaliplatin inhibited the leukemia cell growth in concentration- and time-dependent manners.Marked cells accumulated in G_2/M phase.The rate of cell growth inhibition was 85% after intervention in 0.016 mmol/L oxaliplatin for 72 hours.The cells showed G_2/M arrest and apoptosis.Conclusion:Oxaliplatin suppresses the growth of K562 cells in vitro by causing cell-cycle arrest,and cell apoptosis.The mechanisms of oxaliplatin provide theoretical guidance for clinical treatment.
