Effects of combination of arsenic trioxide and octreotide on proliferation and cellular apoptosis in human hepatocellular carcinoma

Abstract:
Objective:To study the effects of combination of arsenic trioxide and octreotide on the proliferation and apoptosis of hepatocellular carcinoma cells(HCC),and to explore the mechanism of their action on liver cancer.Methods:Heptocellular carcinoma cell SMMC-7721 was treated with arsenic trioxide and octreotide.Cell survival rate was determined by MTT assay;cell cycle and the apoptosis index were determined by flow cytometry (FCM);the morphological changes were observed by transmission electron microscope.Results:The combination of arsenic trioxide and octreotide significantly inhibited the growth of SMMC-7721 and the concentration of each drug in combination is 2.022?mol/L and 8.088?mol/L that was less than single use of arsenic trioxide (3.707?mol/L)and octreotide (22.637?mol/L).The combination of two drugs can arrest HCC in S phases and induce apoptosis.Conclusion:Arsenic trioxide can enhance the antiproliferative effect of octreotide to human HCC and induce apoptosis.
