Angiotensin Ⅱ type 1 receptor mRNA and protein expression in human pancreatic cancer cell lines

Abstract:
Objective The incidence of pancreatic cancer appears to be increasing and the prognosis is poor. Recent studies have revealed that angiotensin Ⅱ type 1 receptor (AT1) stimulates the growth and angiogenesis of tumor, and selective AT1 antagonist inhibits these effects. This study is investigated the expression of AT1 in pancreatic cancer cell lines. Methods The pancreatic cancer cell lines used in this study were SW1990, PaTu8988s and PANC-1. RT-PCR was used to detect the AT1 mRNA expression, and ABC immunocellularchemical staining and sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) were used to detect the AT1 protein expression. Results Both AT1 mRNA and protein were expressed in the three cell lines. The AT1 protein was found in the cell membrane and cytoplasm of these cells. The AT1 protein ( M r 44?10 3) was also demonstrated by SDS-PAGE. Conclusion The results suggest AT1 may play an important role in pancreatic cancer growth, and inhibition of AT1 might be a useful therapeutic strategy.
