Abstract:
Objective:A method for the quantitative determination of adenosine phosphates of mouse myocardium by HPLC is described. Methods:A ODS C 18 column and a mobile phase of methol-50mmol?L -1 phosphate (1∶99)buffer solution (pH 6.07),with UV detector at 254nm were used. Results:The average recovery rates of ATP,ADP and AMP were 101.7%?9.3%,94.8%?10.6% and 98.3%?5.7% respectively;the RSDs of within-day were 3.5%,6.8% and 4.8%,and the RSDs of between-days were 6.4%,8.6% and 8.2%,respectively.Conclusion:The method is simple, rapid and senstive and can be used to analyse the adenosine phosphates in myocardium. The myocardial contents of ATP,ADP as well as AMP decline notably in mice with viral myocarditis.
