INHIBITION OF CELL GROWTH OF HUMAN HEPATOMA CELL LINE BEL-7402 BY DIBUTYRYL CYCLIC AMP AND AMINOPHILLINE

Abstract:
It has been well documented that cyclic AMP and its derivative dibutyryl cyclic AMP (dbcAMP) preferably inhibit the growth of transformed fibroblasts in vitro. However, there have been no much information concerning the action of dbcAMP on established human cancer cell line. The present study was designed to examine the effects of dbcAMP, aminophilline and of the combination of these two agents on human hepatoma cell line BEL-7402. In order to study the possible mechanism of the action of dbcAMP, the influence of dbcAMP on the ultrastructure of the cells was observed. The results showed that both dbcAMP (0.2 mM)or aminophilline (0.2 mM) alone could inhibit the growth of the cells and as the concentration increased, the inhibitory effect increased too. The combined use of dbcAMP and aminophilline exerted stronger action on cell growth. However, the effects of either dbcAMP and aminophilline alone, or combined use of two agents were reversible.Microscopic observation showed that the cells became longer and thinner, even with long cytoplasmic processes (Fig 2) after the treatment with dbcAMP plus aminophilline. The most prominent changes of ultrastructures induced by both agents included the folowing three aspects: Ⅰ. microvilli on the cell surface decreased in number (Fig 3,5). 2. tonofibrils increased in number (Fig 4). 3. polyribosomes assembled into irregular masses and lost the clustered structure (Fig 4). Based on the above observation, it might suggest that the inhibitory effect of dbcAMP may be due to its action in inducing a certain degree of differentiation of BEL-7402 cells.
