STUDIES IN VITRO ON THE INDUCTIVE ACTION OF TUMOUR TISSUES ON THE ECTODERM OF CHICK BLASTODERM

Abstract:
It has been demonstrated that some tumour tissues can induce ectoderm of amphibian embryo to differentiate. In order to accumulate informations about inductive action of tumour tissues on the ectoderm of chick blastoderm, several malignant tissues, both fresh or treated with 70% alcohoI, were taken from Rous sarcoma, Walker cancer-sarcoma, sarcoma 180, spindle cell sarcoma and esophageal cancer cell line (109 cell line) as inductor, while the ectoderm was taken from the region lateral to the node and streak of blastoderm of stage 4 of chick embryos. They were brought together by the "Sandwich" technique as described elsewhere (Chang). The explants were then cultivated on the area vasculosa of 1~2 days chick blastoderm for 1~5 days. The specimen were preserved in Bouin's fixitive and stained with hemotoxylin eosin. The results showed that in all of the cases except two the neural formation was not induced in the ectoderm of the blastoderm by the tumour tissues used. The two cases that showed atypical neural plates were induced by the fresh tissue from sarcoma 180 and the tissue from spindle cell sarcoma treated with 70% alcohol. In less than one half of the cases the capacity for placoid thickening response of the ectoderm (Fig 1,2) or atypical epiderm proliferation (Fig 3) was observed, while most of the remaining cases were incapable to show any response to tumour tissues. The possible role of the ectoderm of chick blastoderm in responding to tumor tissues was discussed in relation to that of amphibian embryo.
