Abstract:
Objective To investigate the effects of Nordy on induced differentiation of HPV16 subgenes-immortalized human endocervical cells(H8 cells). Methods After treated with Nordy, the proliferation of H8 was tested with MTT assay. The effect of Nordy on cell cycle of H8 was analyzed by flow cytometer(FCM).The expression of Ki67 of H8 was detected by immunohistochemistry. The activity of telomerase was tested by TRAP-ELISA.Results Nordy could inhibit proliferation of H8 cells significantly.H8 cells were arrested in G_(0)/G_(1) phase after Nordy treatment. The Ki67 expression and telomerase activity reduced significantly after Nordy treatment. Conclusion Nordy could induce the differentiation of H8 cell lines by inhibiting their proliferation, blocking cell cycle and decreasing telomerase activity.
