Abstract:
Objective:To observe the influence of ischemia/reperfusion(simulated by anoxia) on the characteristics of K~(+) channels in renal tubular epithelial cells. Methods: Healthy renal tubule epithelial cells were divided into normal control group(cultured in serum-free RPMI 1640 medium),anoxic group(treated with antimycin A + serum-free RPMI 1640 medium),and oxidative stress group(H_(2)O_(2)+ serum-free RPMI 1640 medium)(n=15).The whole-cell patch clamp technique was used to examine the characteristics of K~(+) channels in renal tubular epithelial cells. Results: The transmembrane K~(+) current in tubular epithelial cells was voltage-dependent and could be blocked by K~(+) channel blocker Ba~(2+).The transmembrane potential levels in anoxic group and oxidative stress group were remarkably higher than that in normal control group(P
