Therapeutical effect of isoflavones on experimental nephrotic syndrome in rats

Abstract:
Objective To investigate the therapeutical effect of isoflavones on rats with adriamycin (ADR)-induced nephrotic syndrome (NS). Methods Thirty-two SD rats were randomly divided into four groups. NS models were induced by tail intravenous injection with ADR(5. 0 mg/kg) in Group A, B and C. Group D was treated with 0. 9% NaCl as normal control. Group A was received soybean protein diet which contained isoflavones 3. 4 mg/g protein. Group B received soybean protein diet without isoflavones. Group C and Group D received casein diet as well. Serum biochemical indexes(TG, Tch, TP, Alb, HDL, LDL, Ca, Fe), urine protein, renal function indexes(BUN, Cr) and renal antioxidant indexes (SOD, MDA, GSH-Px, GSH) were examined. The changes of kidney tissue were observed by light and electron microscopy and the positive rate of TGF-?1 in kidney was examined by immunohistochemistry. Results Hyperlipidemia, hypoalbuminemia and proteinuria were observed in Group A, B and C. In these 3 groups, TP, Alb decreased and TG, Tch increased as compared with those in Group D . In Group A, Tch and TG were lower than those in Group C. Injuries of renal function were observed in all the NS rats. But BUN in Group A was significanlly lower than that in Group C. There was no markedly differences of SOD and GSH between all groups. GSH-Px in Group A was higher and MDA was lower than that in Group C . Morphologic findings by electron microscopy noted widespread fusion of foot processes and loss of foot process architecture. Group A was better than Group B and C. The positive rate of TGF-?1 in Group A was lower than those in Group B and C. Conclusion Isoflavones has protective effects on renal structure and function of NS rats.
