Preliminary Observation on 4-dimethylaminophenol Intoxication in Human Beings

Abstract:
4-dimethylaminophenol hydrochloride (4-DMAP) was injected intramucularly to 5 healthy individuals(4 male and 1 female) in the dose of 3 mg/kg. The mean concentration of methemoglobin (MHb) increased to 25% of the total hemoglobin in 30 minutes after injection. The HT-value was 9 minutes. 3 hours after injection, the MHb level decreased to 17% of the total hemoglobin, then it continued to decrease but in a slower rate.All the 5 individuals became cyanotic 5 minutes after injection-as the MHb level was increasing. There was obvious and localized tension pains on the site of injection 20-60 minutes after injection. All the 5 complained severe but tolerable headache 80 minutes after injection. 12 hours after injection, there was low-grade fever and elevation of white blood cell count, which returned to normal in the following day. On the second day of injection, they complained of uneasiness in walking because of the pains of the injected thigh and sense of fatigue. The pains over the injection si te subsided one week after injection. The functions of the liver, the kidneys and the cardiovascular system showed nothing abnormal.
