Abstract:
Hemoglobin J Bangkok is a fast-moving ? chain variant of hemoglobin (Hb) found in a patient living in Baoshan district, the suburb of Shanghai.This abnormal Hb comprised 50.1% of total Hb of the pateint. No remarkable changes were detected in hematological examinations of the patient's peripheral blood.The iso-propanol precipitation test was negative and Heinz bodies were not found.From the pedigree study it was found that the grandfather and father of the propositus were also carriers of the same abnormal Hb.The abnormal ? chain was separated by PCMB and CM-22 cellulose column chromatography and was then digested with TPCK for analysis of finger-prints and determination of the amino acid composition and sequence of the abnormal pep-tide. Thus the glycine residue at the position 5G on ? chain(?56[DT]) was replaced by aspartic acid,and it was hence identified as hemoglobin J Bangkok(?2,?256[DT]Gly→ASP).
