Effect of retinoic acid on the expression of NDRG4,a human brain/heart-specific gene,in NT2 neuronal precursor cells

Abstract:
Objective To investigate the effect of retinoic acid(RA) on the expression of NDRG4, a gene specifically expressed in human brain and heart, in human neuronal precursor cell line NT2. Methods NT2 cells were cultured and recovered before and 1 or 2 weeks after being treated with retinoic acid. Total RNA was prepared and subjected to Northern blot analysis. Reverse transcription-polymerase chain reaction(RT-PCR) was also performed using the primers spanning the 5'-region specific to NDRG4-H or NDRG4-B variant cDNAs, respectively, and the primer spanning the common region shared by both variant cDNAs. Results The expression of NDRG4 was significantly higher in NT2 cells treated by RA than in NT2 untreated cells. NDRG4-B and NDRG4-H demonstrated different responses to RA treatment. The expression of NDRG4-H was increased, whereas the expression of NDRG4-B was not changed by RA treatment. Conclusions NDRG4 may be related to the development and growth of the neurons of central nervous system. Further study of NDRG4 might be beneficial for understanding the pathogenesis of neurodegenerative disorders, such as Alzheimer's disease, etc.
