Abstract:
Objective:To study in vivo antigenic modulation of human T lymphocytes induced by OKT3.Methods:We monitored T lymphocytes and subsets of 19 OKT3 treated kindey graft recipients by three color flow cytometry.T cell counts and fluorescent intensity were done on the day before the first OKT3 injection, one day, three days and five days during OKT3 injection, one day and five days after OKT3 injection. Results:The percentage of CD3 +,CD3 +CD4 + and CD3 +CD8 + cells were dramatically decreased and disappeared following the first injection of OKT3, and the fluorescent intensity of the CD3 + cells were also significantly decreased 86%,while modulating CD3 -CD4 + and CD3 -CD8 + cells were observed but were functionally inactive. After OKT3 was stopped, CD3 +,CD3 +CD4 + and CD3 +CD8 + cells were gradually increased, the fluorescent intensity of the CD3 + cells began to evaluate, while CD3 -CD4 + and CD3 -CD8 + cells were decreased and vanished. Conclusion:Antigenic modulation of human T lymphocytes by OKT3 results in an appearance of modulating cells,it suggests that accurately monitoring CD4/CD8 should be done in transplant rejection.
