Study on the Distribution of Moxifloxacin in Ocular Tissues and Its Pharmacokinetics in Rabbits with Intravenous and Perfusion Administration

Abstract:
OBJECTIVE: To compare the distribution of moxifloxacin in ocular tissue and its pharmacokinetics in rabbit with intravenous and perfusion administration. METHODS: 54 rabbits were divided into intravenous administration group and perfusion administration group (n=27 each). Each group were randomized into 9 sub-groups (n=3). Those groups were given 24 mg?kg-1 moxifloxacin with corresponding adminstration and HPLC was used to detect plasma concentration of moxifloxacin in ocular tissue. 3p97 software was adopted to calculate pharmacokinetic parameters. RESULTS: The Cmax of moxifloxacin by perfusion administration vs. intravenous administration in cornea,aqueous humor, iris-ciliary body,lens and vitreous were (7.09?4.708) ?g?g-1 vs. (2.55?0.382) ?g?g-1; (3.84?1.217) ?g?g-1 vs. (3.71?0.54) ?g?g-1; (5.87?1.93) ?g?g-1 vs. (2.72?0.662) ?g?g-1; (1.36?0.176) ?g?g-1 vs. (0.74?0.396) ?g?g-1; (1.87?0.586) ?g?g-1 vs. (0.87?0.073) ?g?g-1. The AUC0~t of two groups in above tissues were (12.24?1.112) ?g?h?g-1 vs. (9.27?0.559) ?g?h?g-1; (4.71?0.082) ?g?h?g-1 vs. (2.33?0.399) ?g?h?g-1; (13.79?0.814) ?g?h?g-1 vs. (8.04?2.045) ?g?h?g-1; (5.83?0.309) ?g?h?g-1 vs. (3.61?0.41) ?g?h?g-1; (3.45?0.064) ?g?h?g-1 vs. (3.09?0.539) ?g?h?g-1. The pharmacokinetic parameters in perfusion administration group were all higher than that in intravenous administration group (P
