Clinical significance of increase in mitochondrial DNA copy number in laryngeal squamous cell carcinoma

Abstract:
OBJECTIVE To study the relationship between mitochondrial DNA copy numbers abnormality,and theTNM and differentiation of laryngeal squamous cell carcinoma. METHODS Total DNA was extracted from microdissected cancerous and pericancerous tissues from 32 laryngeal squamous cell carcinoma samples. Using the SYBR GreenⅠquantitative polymerase chain reaction,the mitochondrial to nuclear DNA ratio was determined by quantification of ND1 and ?-actin. RESULTS Mean ?-actin/ND1 DNA ratios for pericancerous and cancerous tissues of LSCC in early stage (ⅠandⅡ) LSCC tissues were 0.269 and 0.274 respectively,and were 0.168 and 0.157 respectively in advanced staged (Ⅲ and Ⅳ) LSCC tissues. The mean ?-actin/ND1 DNA ratios for pericancerous tissues from poorly,moderately and well differentiated samples were 0.184、0.237 and 0.239 respectively,and were 0.175,0.201 and 0.226 respectively for cancerous tissues. CONCLUSION The mitochondrial DNA copy number of advanced staged(Ⅲ and Ⅳ) LSCC samples were significantly higher than that of early staged(ⅠandⅡ) LSCC samples. mtDNA copy numbers were inversely correlated with the degree of differentiation,sugges-ting that increases in mtDNA copy numbers may be an early event in the pathogenesis of LSCC.
