Abstract:
OBJECTIVE:To establish the method of determining the gliclazide in human serum by solid-phase extraction combined with HPLC.METHODS:The pre-treatment of plasma sample was carried out by solid-phase extraction.The chromatographic column was Zirchrom;the mobile phase was methyl cyanide-0.2%glacial acetic acid(60∶40);the UV de-tection wavelength was229nm;the column temperature was40℃and the flow rate was1.0ml/min.The blood concentration of gliclazide was determined with glipizide as the internal standard.RESULTS:The calibration curve was linear within the range of0.1～10.0?g/ml(r=0.9966);the average recovery was99.96%;the RSD of both intraday and inter-day were below10%. CONCLUSION:The method is simple and the determination result is accurate and with little interference,which is suitable for the study of the pharmacokinetics and bioavailability of gliclazide.
