Morphologic Observation and Biological Behavior Research of Rabbit periosteal Osteoblasts Incubated in Vitro

Abstract:
We developed a culture system of osteoblasts isolated from juvenile rabbit periosteum. Having referred to certain reports about osteoblasts qualifying method, authors have performed an experimental study on morpology, histochemistry and biological properties of cultured cells. This study confirmed that the cultured cells are typical osteoblasts characteristics of:(a). Under the light microscope, the cultured cells conformed to osteoblasts morphologically; (b). Under the electron microscope, their ultrastructure was similar to those of osteoblasts——sunken muclei, dilatation of rough endoplasmic reticulum, a great deal of mitochondrion, secretory vacuole and electron dence substance in cytoplasm; (c). Rabbit periosteal osteoblasts in culture still maintain high AKP (alkaline phosphatase) activities;(d). The cultured cells have certain capacity of calcification in vitro.The cultivation could be used biologically to study characte ristics of bone metabolism, to probe into osteogenic capacity of implanting cells in vivo. Osteoblastic studies could, clinically and experimentally, be based on the culture system described in this paper.
