Effect of propofol on autophagy during ischemia-reperfusion injury to rat hearts

Abstract:
Objective To observe the effects of propofol on mTOR and p-mTOR expression during myocardial ischemia-reperfusion (I/R)injury in type 2 diabetic rats.Methods A total of 60 male SD rats were randomly divided into six equal groups as follows (n = 7 ):the sham operation group (CC),who underwent sham operation without tighting the coronary artery sutures;myocardial ischemia-reperfusion group (CI),who was induced by occluding the left anterior descending coronary artery for 30 min,followed by 2 h of reperfusion;the myocardial ischemia-reperfusion+propofol group (CI+P),who underwent I/R and propofol infusion at 6 mg·kg-1 ·h-1 starting 10 min before oc-cluding the left anterior descending coronary artery;the diabetic sham operation group (DC),diabetic I/R group (DI)and diabetic propofol group (DI+P),in which the procedures mentioned above were applied to the rats with type 2 diabetic.Type 2 diabetic rats were induced by a combination of high-fat and high-glucose diet intraperitoneal STZ injection.Before tighting the coronary artery and after 30 min of tighting the coronary artery and after 2 h of reperfusion,the HR,LVSP and ±dp/dtmax after 120 min of reperfusion were recorded.The serum concentrations of nitric oxide (NO)and endothelin (ET-1 )was measured,the injured cardiac tissues were removed for examing the expression of mTOR,p-mTOR.Results There were no signifcant differences in the parameters of hemodynamy between the groups before tighting of the coronary artery.Compared with group CC,no significant change was found in the parameters of hemodynamies in group DC,but the HR,LVSP and ± dp/dtmax was significantly decreased in other groups (P <0.05).Compared with group CC,the serum con-centration of ET-1 after 2 h of reperfusion was increased in the groups CI and DI,while the concentra-tion of NO in these groups decreased after reperfusion (P <0.05 ).Compared with the I/R groups, propofol increased the content of NO and decreased the content of ET-1 (P <0.05).Compared with the sham operation groups,the I/R groups decreased the expression of p-mTOR,increased the ex-pression of mTOR;while compared with the I/R groups,propofol increased the expression of p-mTOR and decreased the expression of mTOR (P < 0.05 ).Conclusion These data suggest that propofol can prevent myocardial ischemia-reperfusion injury in both normal and type 2 diabetic rats, possibly by attenuating endothelial cell injury and inhibiting the autophagy of cardiomyocytes.
