Effects of QingReYangYin Recipe on the Ovaryhormone and Local Insulin Resistance in Rat Model of Polycystic Ovary Syndrome

Abstract:
Objective]To observe and understand the effect and mechanism of QingReYangYin Recipe(QRYYR) on the ovary hormone level and mRNA expressions of PI3K and PKB in adipose tissue around womb and ovary in rat model of polycystic ovary syndrome(PCOS).[Methods]Rat PCOS model was established by Poretsky method and divided randomly into four groups: model group, western medicine group,low dose QRYYR group and high dose QRYYR group. Normal rats were used as normal group. All rats were orally administrated for 14 days and were weighed every day.After 14 days treatment, serum sample of each rat was obtained for analysis of estradiol(E2),progesterone(P), testosterone(T) contents. Each ovary was collected and weighed for measurement of the ovarian index. The fat pad around womb and ovary was obtained for analyzing the mRNA expressions of PI3K and PKB by PCR. [Results]Compared with the model group, the serum E2 and P levels in QRYYR group were significantly higher but T level was significantly lower(P<0.05) than the model group. The levels of E2, P and T in high dose QRYYR group had no difference from that in the western medicine group and the normal group(P>0.05).The ovarian index in the QRYYR groups was significantly decreased as compared with the model group(P<0.05),and that in the high dose QRYYR groups had no difference with the normal group(P>0.05).The mRNA expressions of PI3K and PKB were significantly increased in the QRYYR groups as compared with the model group(P<0.05),and the expressions in high dose QRYYR group, western medicine group and the normal group had no significant difference from each other(P>0.05). [Conclusion] QRYYR could significantly increase the content of E2 and P in the rat serum, significantly decrease the content of T and the ovarian index. Meanwhile, it could significantly increase the mRNA expressions of PI3K and PKB in adipose tissue. It could improve the ovarian function and decrease the local insulin resistance. The effect of the high dosage group on PCOS was superior or similar to Metformin.
