Expression of ADM and ADM mRNA in the cerebral cortex after focal cerebral ischemia/reperfusion in rats

Abstract:
Objective To discuss the relationship between ADM and the injury of focal cerebral ischemia/reperfusion. Methods Construct the model of the rat middle cerebral artery ischemia/reperfusion by the line-block-method which the reperfusion is applied after the ischemia lasting for 2 h. By the method of immunohistochemistry and RT-PCR, we test the expression of ADM and ADM mRNA after focal cerebral ischemia/reperfusion in the brain of rats in different time, and make a dynamic observation at the same time. Results ADM and ADM mRNA express in the normal rat's brain, but lower than the sham operated group, P>0.05. There is a over expression of ADM and ADM mRNA after focal cerebral ischemia/reperfusion in the brain of rats, the difference is obviously compared with the normal group and the sham operated group, P<0.05. After the ischemia last for 2 h, the dynamic observation showed that ADM and ADM mRNA began to express in the cerebral cortex when the reperfusion last for 2 h after, and reach the peak time at 22 h, and still more than the normal group at the time of 1 w, P<0.05. Conclusion ADM and ADM mRNA showed a regularitily over expression after ischemia/reperfusion of the brain.
