Abstract:
Objective:To study the effect of Src kinase inhibitor PP2 on learning and memory in rats with chronic alcohol(EtOH)exposure and its molecular mechanism.Methods:The chronic alcohol exposure model of rats was pre-pared by drinking method,and PP2 was given by intraperitoneal injection.The learning and memory ability of rats was detected by Morris water maze,the contents of IL-1β and TNF-α in hippocampus were detected by enzyme linked im-munosorbent assay(ELISA),and the expressions of Src,Iba-1 and iNOS in hippocampus were detected by Western Blot.Results:Chronic alcohol exposure could prolong the incubation period and escape time of rats.At the same time,the expression of Src,Iba-1 and iNOS,as well as the contents of IL-1β and TNF-α were increased in the hippocampus.PP2 can decrease the escape latency and the expression of Src,Iba-1,and iNOS in hippocampus of chronic alcohol exposed rats.In addition,the levels of IL-1β and TNF-α decreased.Conclusion:PP2 improves learning and memory impairment caused by chronic alcohol exposure by inhibiting nervous system inflammation.
