Abstract:
Objective To compare the efficacy of alleviating the symptoms of menopause by application of different dosage of estradiol by percutaneous (0. 75 mg vs 1. 5 mg) combined with progestogen via oral. Methods A total of 60 patients with postmenopausal women were randomly divided into 4 groups, received different dosage: 17 beta estradiol (E2) by percutaneous, material progesterone ( MP) and medroxyprogesterone acetate (MPA) , each cycle lasted 25d. Adverse effects, such as menopausal symptoms and irregular vaginal bleeding were observed. Results All symptoms of menopause has improved ( P < 0. 01 ), the differences between groups were not significant ( P > 0. 05 ). The symptoms, such as hot flashes, sleep and mood symptoms improved quickly, while the following: tired, inattention, lack of libido improved slowly. The incidence of irregular vaginal bleeding among MP 100mg/d group showed lower than that in 2mg/d MPA group (P <0. 01 ). Conclusions Two long-term continuous use of estradiol (0.75 mg vs 1.5 mg) combined with progestogen both can effectively alleviate menopausal symptoms, and the symptoms of hot flashes, sleep and mood improved earlier. Combining with 100mg/d MP is superior to avoid irregular vaginal bleeding than with 2mg MPA. Thus clinical application should be individualized.
