Factors affecting release of adrenomedullin and adrenotensin from the aortae of rats

Abstract:
Objective To investigate the release regulation of adrenomedullin (ADM) and adrenotensin (ADT).Methods Incubation or perfusion of the isolated aortae of rats for 1 hr with different factors and radioimmunoassay for ADM and ADT in the media.69%-300% in a dose-dependent manner. One and 10 nmol/L AⅡ and ET-1 had no obvious effects on ADT release. 100 nmol/L AⅡ and ET-1 increased ADT release by 128% and 117%, respectively. The ratios of ADM/ADT of the above-mentioned groups were increased by 36%-111%. 10 nmol/L epinephrine and calcitonin gene related peptide (CGRP) augmented release of ADM and ADT by 101% and 87% and decreased their release by42% and 42%, respectively. However, the ratios of ADM/ADT of the two groups were unchanged. Hypoxia had no effect on ADM release, but it inhibited ADT release by 43%. The ratio of ADM/ADT was increased by 87%. 1 μg/ml lipopolysaccharide (LPS) had no effect on release of ADM and ADT, but 5 μg/ml and 10 μg/ml LPS increased ADM release by 177% and 239%. They increased ADT release by 39% and 74% and ADM/ADT by 97% and 98%, respectively.*h-1, 0.252±0.060 pmol/gw*h-1 and 0.844±0.115, respectively.
