Effects of Gender and Age on the Expression of Interferon Stimulated Gene in the Peripheral Blood of Patients with Chronic Hepatitis C

Abstract:
Objective:To investigate the mRNA expression of interferon‐induced protein with tetratricopeptide repeats 1 (IFIT1) and IFIT3 in the peripheral blood mononuclear cells (PBMC) in patients with chronic hepatitis C (CHC) with different genders and ages .Methods :Anticoagulant whole blood was collected from 215 patients with CHC and 79 healthy individuals . The mRNA expression levels of IFIT1 and IFIT3 were detected by real‐time quantitative PCR .Alanine aminotransferase(ALT) and aspartate aminotransferase (AST ) were detected by automatic biochemical analyzer . HCV‐RNA was detected with fluorescent probe .Results:Expression levels of IFIT1 and IFIT3 in PBMC of patients with CHC were significantly different from those in healthy people (t=22 .35 , P< 0 .05 ;t= 22 .75 , P< 0 .05) .There were statistically significant differences in ALT and AST between patients with CHC and healthy people (t=34 .38 , P<0 .05 ;t=21 .38 , P<0 .05) .The correlation analysis of gene expression levels of IFIT 1 and IFIT3 in PBMC with age ,gender ,liver function and HCV load in CHC patients revealed that the mRNA expression levels of IFIT 1 and IFIT3 were positively correlated with age (r=0 .395 , P<0 .05 ;r=0 .482 ,P<0 .05) and gender(r=0 .453 ,P<0 .05 ;r=0 .358 ,P<0 .05) .Conclusions :The expressions of IFIT1 and IFIT3 in PBMC are correlated with gender and age in CHC patients .
