Abstract:
Objective:To clarify the infection status and gene characteristics of human herpesvirus 6 (HHV-6) in exanthema subitum cases in Gansu province in 2011.Methods:Throat swabs collected from 169 cases with exanthema subitum in Gansu province in 2011 were tested by the quantitative real-time PCR method. PCR amplification and sequence determination for U90 gene were performed for HHV-6 nucleic acid positive specimens and then the characteristics of U90 gene were analyzed in this study. Results:Sixty out of 169 samples from exanthema subitum cases were positive for HHV-6 nucleic acid, and 10 sequences of HHV-6B U90 gene were obtained. HHV-6 positive cases were mainly concentrated in infants under 2 years of age, with a median age of 9 months. Phylogenetic analysis showed that the sequences of HHV-6B could be divided into clades a and b. And then the clade a could be further divided into three lineages (lineage a-1, a-2 and a-3) with geographical clustering. The 10 HHV-6B sequences obtained in this study were scattered in different branches of the phylogenetic tree, belonging to lineages a-1 and a-2, and clade b. No gene recombination was found in these 10 sequences. Conclusions:Multiple HHV-6B clades/lineages were circulating in Gansu province, and the virus strains showed genetic polymorphism. Through this study, the gene characteristics of HHV-6B circulating in Gansu province were firstly clarified, which not only filled the gap of the HHV-6B molecular epidemiology in China, but also accumulated the data for global HHV-6B research.
