Abstract:
Objective:To study the effect of mixed sample detection in the diagnosis of COVID-19, to find the best method of the detection, and to provide scientific data for prevention and control of COVID-19.Methods:We Chose the positive samples with different Ct values, mixed with the following numbers of negative samples: 1, 2, 5, 10, 20, 30, 40, 50 and 100 and pooled, and then detected the Ct values of mixed samples by real-time RT-PCR.We analyzed the influence of mixed detection on experimental result , and found the best mixed detection method . Results:After the mixed samples were detected, the positive samples with Ct<30 could be detected in 100 samples, the positive samples with Ct between 30 and 35 could not be detected in 50 samples, and the samples with Ct>35 could not be detected in 5 samples. Conclusions:The method of "single sampling and mixed testing" should be adopted in the mixed sample detection, and no more than 5 samples should be sampled and mixed by volume, which can effectively improve the detection efficiency and reduce the rate of missed detection.
