Establishing orthotopic and metastatic colon cancer model by submucous injection of HCT116 cells in nude mice

Abstract:
Objective To establish a simple,feasible and reliable orthotopic implantation and metastasis model of human colon cancer in nude mice cecum mucosa.Methods Tumor cell line HCT116,derived from human colon cancer,was injected into colon cecum mucosa of nude mice to develop colorectal cancer model.HE staining and human carcinoma antigen (CEA) immunohistochemistry were performed to detect tumor in situ and metastasis condition in abdominal lymph nodes,the liver and the lungs.Results On the 2nd week after inoculation,90% of the nude mice developed cecum tumor of 2-5 mm in diameter in situ.Abdominal lymph node metastasis was observed on the 4th week and present in all mice at week 12.Liver and lung metastasis started to appear at a percentage of 60% on the 10th week and 39% on the 12th week.Conclusion Human colon cancer in situ could be successfully established by inoculating HCT116 into cecum mucosa of nude mice to mimic the clinical development and growth of colon cancer.This method proves to be simple with a high tumor-bearing rate.It could be used as a reliable and ootimal animal model for in-depth basic research about the mechanism of colon cancer metastasis.
