Abstract:
Objective To analyze the characteristic of EBER gene of Epstein—Barr virus in EBV infection in children in Beijing area.Methods Total DNA was extracted from whole blood of 288 patents with EBV-associated infectious mononucleosis (IM),52 patients with EBV-associated hemophagocytic lymphohistiocytosis (EBV-HLH) and 9 patients with chronic active Epstein-Barr virus (CAEBV) infection in Beijing Children' s Hospital.The subtype of EBV type Ⅰ / Ⅱ was defined by the size of amplicon of the EBNA3C genes.The amplified EBER gene products were sequenced directly and the sequences were analyzed by BioEdit 7.0.9 and MEGA 4.0.2.The relationship between EBV subtype or EBER genotype and clinical phenotype were statistically analyzed.Results Type Ⅰ EBV was present in 98.6% samples (344/ 349) and type Ⅱ EBV in 1.4% samples (5/349).Three EBER genotypes,including EB-6m,EB-8m and EB-7m,were defined in this study.Thc detection rates of EB-6m,EB-8m and EB-7m were 96%,2.9% and 1.1% respectively.There were no significant differences in the frequencies of the three genotypes among IM,EBV-HLH and CAEBV (P > 0.05).Conclusion EBV type Ⅰ is predominant in EBV infection in children in Beijing area.EB-6m genotype was predominant genotype in this area.
