Effect and regulation of Zuogui Decoction on expression of ovarian P450 aromatase in rats during perimenopausal period

Abstract:
Objective To study the effect and regulation of Zuogui Decoction on the expressions of ovarian P450 aromatase (P450arom) and CYP19-mRNA in rats during perimenopausal period.Methods The rat model of perimenopausal period was respectively given Zuogui Decoction in high, middle and low dose orally for 8 weeks. The serum content of estradiol (E_2) was detected by using radioimmunoassay (RIA), and the expressions of ovarian CYP19-mRNA and P450arom were detected by applying RT-PCR and immunohistochemistry. The granulocytes were collected and cultured from immature rats after injection of pregnant mare serum. The expression of CYP19-mRNA with different treatments was detected by using RT-PCR. Results The serum level of E_2 and the expressions of P450arom and CYP19 were significantly lower in the perimenopausal period group than those in the young group (P<0.01), while those increased significantly (P<0.01) after the treatment with mid-dose and high-dose Zuogui Decoction. The outcomes of different treatments in cultured granulocytes showed that genistein, forskolin and extract of Zuogui Decoction improved the expression of CYP19-mRNA (P<0.05) compared with the control group. The extract of Zuogui Decoction combined with phorbol ester (PMA) or genistein improved further the expression of CYP19-mRNA (P<0.05) compared with extract of Zuogui Decoction.Conclusion The mid-dose and high-dose Zuogui Decoction can increase the serum content of E_2 through up-regulating the expressions of CYP19 and P450arom, and it also can improve the expression of CYP19-mRNA through activating PKC pathway or promoting the action of genistein in rats during perimenopausal period.
