Interventional effect of Shugantongluo Formula on cyclomastopathy in rats

Abstract:
Objective To observe the effect of Shugantongluo Formula on eyclomagtopathy in mts and the mechanism.Methods All unpregnancy female Wistar rats(n=84)were randomly divided into the blank control group(group 1),model group(group 2),tamoxifen group(group 3),Rupixiao group(group 4),and high-dose,mid-dose and low-dose Shugantongluo Formula group(group 5,6 and 7,each n=12),The rat model of eyclomastopathy was established by injecting benzoate estradiol and progestogen intramuscularly for 30 days.The histomorphological changes of mammary glands,uterus and ovary,and the quantity and diameter of mammary lobule and alveolus were observed.The serum levels of estradiol(E_2),progesterone(P),prolaetin(PRL),luteal hormone(LH),follicle-stimulating hormone(FSH),testosterone(T),and estrogen receptor(ER)and progesterone receptor(PR)in mammary glands were detected.Results In group 5,6 and 7 the quantity of mammary lobule,and quantity and diameter of alveolus were reduced(P＜0.01)and the serum E_2 level was decreased with a significant difference compared with group 2 (P ＜ 0. 01). In group 5 and 6 the level of P was increased with a significant difference compared with group 2 (P ＜0.05). In group 5 the level of PBL was decreased with a significant difference compared with group 2 (P ＜ 0. 05 ). The levels of FSH, LH and T had no significant changes without difference compared with group 2 (P＞0.05 ). In group 5 and 6 the expressions of ER and PR were regulated downward, which values was different significantly compared with those in group 2 ( P ＜ 0. 05,P ＜ 0. 01 ) and had no difference compared with those in group 3 ( P＞0. 05 ). Shugantongluo Formula inhibited the paraplasm of uterus and ovary tissue. Conclusion Shugantongluo Formula has a therapeutic effect on cyclomastopathy in rats, and the mechanism may be related to that it can correct the disorder of endocrine hormones, and inhibit the higher expressions of ER and PR in mammary glands.
