Influence of Chinese herbal remedies with actions of clearing away heat to remove toxin combined with Jugeng Decoction on TLR4 mRNA expression in rat model of acute lung injury

Abstract:
Objective To observe the influence of Chinese herbal remedies (CHR) with the actions of clearing away heat to remove toxin combined with Jugeng Decoction on the expression of lung toll-like receptor 4 (TLR4) mRNA in rat model of acute lung injury (ALI) induced by endotoxin, and to discuss the synergy mechanism of Jugeng Decoction. Method All male SD rats were divided into the normal saline group, ALI model group, CHR group and CHR combined with Jugeng Decoction group according to the table of random number. The rat model of ALI was established with the femoral venous injection of lipopolysaccharide (LPS). The serum contents of tumor necrosis factor-α (TNF-α) and interleukin-10 (IL-10), the activity of lung myeloperoxidase (MPO) and the expression of TLR4 mRNA were detected in all groups. Result In the ALI model group the serum contents of TNF-α and IL-10 were higher significantly than those in the normal saline group ( P < 0.01 or P <0.05), and MPO activity and TLR4 mRNA expression were enhanced significantly compared with those in the normal saline group ( P <0.01 or P <0.05). After the intervention of CHR the serum contents of TNF-α and IL-10, MPO activity and TLR4 〖JP3〗mRNA expression decreased significantly ( P <0.01 or P <0.05), and the curative effect of the CHR combined with Jugeng Decoction group was better than that of the CHR group ( P <0.05). Conclusion Jugeng Decoction can improve the effect of CHR with the actions of clearing away heat to remove toxin in the reduction of serum contents of TNF-α and IL-10 and inhibition of MPO activity and TLR4 mRNA expression.
