Abstract:
Objective To examine gene expression alteration in fibroblasts of experimental oral leukoplakia.Methods Gene expression profile of fibroblasts from 4-nitroquinoline-1-oxide-induced rat tongue leukoplakia was examined by GeneChip(R) Rat Genome 230 2.0 Array ( Rat 230 2.0) and compared with normal fibroblasts.Then Rat Cytokine Array was applied to analyze 90 cytokines of the early stage ( 8 h) of Nemosis.Results There were 249 genes that were up-regulated in the fibroblasts of leukoplakia and 12 cytokines that were up-regulated in Nemosis.Interleukin (IL)-1 α was the only one which was 2.29 fold up-regulated in Nemosis and kept strengthening in leukoplakia.Conclusions IL-1α from activated fibroblasts may play an important role in the early stage of oral carcinogenesis.It may be a potential target in the early perturbation and treatment of oral premalignant lesions such as leukoplakia.
