Abstract:
Objective To evaluate the biological effects of transforming growth factor-β (TGF-β) on human periodontal ligament fibroblasts (HPDLFs). Methods HPDLFs were primary cultured and examined for the effects of TGF-β on its proliferation, alkaline phosphatase (ALP) activity, osteocalcin (OC) synthesis and formation of the mineralized nodules. Results TGF-β(0.005 0~0.100 0 mg/L) significantly stimulated the proliferation of HPDLFs. The ALP activity of HPDLFs was elevated evidently by 0.005 0 mg/ L TGF-β. TGF-β (0.000 5~0.100 0 mg/L) had no effects on OC synthesis and formation of the mineralized nodules of HPDLFs. Conclusions The effects of TGF-β on HPDLFs are dose-dependent. TGF-β can stimulate HPDLFs to express some of osteoblastic phenotype, and it lacks the ablility to promote maturation of the osteogenic phenotype.
