Expression and clinical significance of B -cell -specific Moloney leukemia virus insertion site-1, proto-oncogene and deleted in liver cancer-1 in cervical cancer

Abstract:
Objective To study the expression and significance of B-cell-specific Moloney leukemia virus insertion site -1 ( BMI-1), proto-oncogene (C-myc) and deleted in liver cancer -1 ( DLC-1) in cervical cancer.Methods A total of 43 cases of cervical cancer were collected as group A , 21 cases of cervical intraepithelial neoplasia as group B and 20 cases of benign lesions as group C .The expression of BMI -1, C -myc and DLC -1 protein was detected by immuno-histochemical PV-9000 two step method.The expression of BMI -1, C-myc and DLC-1 mRNA was detected by real time polymerase chain reaction (RT-PCR), and the relationship with the clinicopathological factors of cervical cancer and the correlation between them were ana-lyzed.Results The expression levels of BMI -1 mRNA in group A, group B and group C were respectively (0.93 ±0.09), (0.29 ±0.06) and (0.17 ±0.03), the difference was statistically significant (P<0.05).The expression levels of C -myc mRNA in group A, group B and C group were (0.96 ±0.08), (0.29 ±0.04) and (0.16 ±0.03), and the difference was sta-tistically significant (P<0.05).The expression levels of DLC -1 mRNA in group A, group B and group C were respec-tively (0.11 ±0.01), (0.83 ±0.11 ) and (0.95 ±0.16 ), the difference was statistically significant ( P <0.05 ) . There was no significant difference in the expression of BMI-1, C-myc and DLC-1 in the tissues of cervical cancer with the age and clinicopathological types of the patients (all P>0.05).There were significant differences in the clini-cal staging, lymph node metastasis and diffe-rentiation of FIGO (all P<0.05).The positive expression rate of BMI -1 and C-myc increased with the clinical stage and differentiation degree of tumor .The positive expression rate of DLC-1 gradually decreased along with the clinical stage and differentiation degree of tumor .There was a negative correlation between the expression of DLC -1 and the expression of BMI -1 and C -myc protein ( all P<0.01 ). Conclusion BMI-1 and C-myc were highly expressed in cervical carcinoma and low expression of DLC -1,all of them are closely related to FIGO stage , the degree of differentiation and lymph node metastasis of cervical cancer pa-tients.The combined detection of BMI -1, C-myc and DLC-1 can be used as a reference index to evaluate the biological behavior and prognosis of cervical cancer .
