Abstract:
Objective To study the effect of Porphyra polysaccharide on blood -brain barrier after cerebral ischemia -reperfusion in mice. Methods A total of 180 male ICR mice were randomly divided into sham operation group , model group, control group and low, middle and high dose experimental groups , 30 mice in each group. The transient middle cerebral artery occlusion ( tMCAO) model was established by the method of suture thread in model group , control group and experimental groups. The sham operation group received similar surgical treatment but no line thrombectomy was performed .Control group was intraperitoneally injected 4 mg· kg -1nimodipine before modeling , low, middle and high dose experimental groups were intraperitoneally injected 100, 200, 400 mg· kg -1Porphyra polysaccharide , once a day, continuously for 7 d. Sham group and model group were intraperitoneally injected with normal saline. The cerebral infarct size and brain wa-ter content of the mice in each group were observed .The permeability of the blood -brain barrier was observed by Evans blue ( EB) method. The expressions of matrix metalloproteinase -9 ( MMP -9 ) and zonula occludens -1 (ZO-1) protein in ischemic cerebral cortex were detected by Western blot .Results The volume of cerebral infarc-tion in model group and low, middle, high dose experimental groups were (28.45 ±5.32 )%, (24.73 ±3.65 )%, (22.36 ±4.19 )%, ( 18.61 ±5.72 )%; brain water content were ( 83.23 ±1.56 )%, ( 80.72 ±1.17 )%, (80.38 ±1.32)%, (79.59 ±1.41)%; brain EB content were (8.02 ±0.71), (7.18 ±0.66), (4.75 ±0.73) and (3.93 ±0.64 ) μg· g-1, the levels of MMP-9 protein in ischemic cerebral cortex were 0.65 ±0.09 , 0.52 ±0.11 , 0.39 ±0.08 and 0.31 ±0.07 ; the levels of ZO -1 protein in cerebral cortex were 0.18 ±0.07 , 0.24 ±0.06 , 0.37 ±0.08 and 0.51 ±0.10 , all with significant differences ( all P<0.05 ).Conclusion Porphyra polysaccharide can down-regulate the expression of MMP -9 in brain tissue after cerebral ischemia -reperfusion, up-regulate the expression of ZO -1 and reduce the permeability of blood -brain barrier, which may be one of the mechanisms of Porphyra polysaccharides protecting brain injury after ischemia -reperfusion.
