Abstract:
Objective To explore the mechanism of curcumol induces apoptosis in human breast cancer cell line michigan cancer foundation-7 (MCF-7) in vitro and in vivo.Methods (1) Cell test:MCF-7 cells were cultured in vitro,blank control group treated with Dulbecco' s modified eagle medium(DMEM),solvent control group treated with 1% alcohol,experimental groups treated with 25,50,100 μmol· L-1 curcumol for 24,48,72 h.Inhibition of cell proliferation was determined by MTT.Cell apoptotic rate was measured by flow cytometry analysis.The expression of B-cell lymphoma-2 (Bcl-2) and Bcl-2-associated Ⅹ protein(Bax) in tumor tissue were examined by RT-polymerase chain reaction(PCR).(2) Animals test:Nude mice were inoculated subcutaneously with MCF-7 cells (1 × 107) to establish the subcutaneous tumor model of human breast cancer,model group treated with normal saline,blank group treated with drug-free liposomes,control group treated with 125 mg · kg-1 elemene,experimental group treated with 100 μmol · kg-1 curcumol liposomes.Tumor-bearing nude mouse were used to study the efficacy of curcumol in vivo.Results (1) Cell test:Curcumol inhibits the proliferation of MCF-7 cells.After 24 h administration,the apoptotic rates in the three concentration experimental groups were (12.80± 2.41)%,(22.10 ± 1.46)% and (33.10 ± 3.25)%,which were significantly different from those in the blank control group (5.36 ± 0.81) % (all P < 0.05).Compared with the blank control group,the expression of Bax was up-regulated and Bcl-2 was down-regulated significantly (all P < 0.05).(2) Animals test:Curcumal can inhibit the growth of tumor in nude mice bearing tumor.Conclusion Curcumol could effectively inhibit proliferation and promote apoptosis of MCF-7 cells,the mechanism may be correlated to decreasing the ratio of Bcl-2/Bax.
