Abstract:
Objective To investigate the roles of icariin combining extracellular matrix metalloproteinase inducer (EMMPRIN) in apoptosis,migration and invasion abilities of human ovarian cancer SKOV3 cells.Methods SKOV3 cells with stably expressed EMMPRIN were established as the test group,SKOV3 cells transfected with pcDNA and SKOV3 cells were the control group and the blank group,respectively.Flow cytometry was performed to measure the apoptosis rate,scratch and Transwell assays were used to detect the migration and invasion abilities.Western blot assay was performed to analyze the expression levels of EMMPRIN,Bcl-2 and Bax.Results The main indexes in blank,control and test groups were compared:apoptosis rates were (10.27 ± 1.35) %,(12.40 ±0.70)% and (5.67 ±0.96)%,average migration indexes were (0.25 ± 0.03),(0.64 ± 0.03) and (O.83 ± 0.02),average number of invasive cells were (45.75 ± 2.87),(120.00 ±3.92) and (153.25 ±8.06),expression of EMMPRIN were (0.12±0.02),(0.22 s±0.01) and (0.34 ±0.02),expression of Bcl-2 were (0.06 ±0.01),(0.55 ±0.02) and (0.87 ±0.03),expression of Bax were (0.37 ± 0.02),(0.28 ± 0.01) and (0.12 ± 0.01).The differences were statistically significant between test group and blank,control groups (P < 0.001,P < 0.01).Conclusion Overexpression of EMMPRIN inhibits apoptosis and promotes migration and invasion abilities of human ovarian cancer SKOV3 cells.
