Effects of Panax Notoginseng decoction on the pharmacokinetics of warfarin in rats

Abstract:
Objective To study the effect of Panax notoginseng decoction (PNS) on the phamacokinetics of warfarin in rats.Methods A total of Sprague Dawley rats were randomly divided into five groups,each group had six rats:warfarin group (0.2 mg · kg-1 · d-1),PNS group (0.54 g · kg-1 · d-1) and combination-L,-M,-H groups [with warfarin 0.2 mg · kg-1 · d-1 and three dosages PNS (0.27,0.54,0.81 g · kg-1 · d-1)].Rats were administrated warfarin once at a fixed time every day,PNS twice per day in the morning and evening,continuous dosing 6 days until reached its steady-state.Blood samples were collected at 0,0.5,1,2,4,8,12,24,36,48,72 h after the last dose on sixth day.Prothrombin time (PT) and International Normalized Ratio (INR) were measured by thromboplastin reagent.The plasma concentration of warfarin was determined by reversed phase-HPLC.Results There is no significant impact on the mean of INR in groups at 0 h.The mean of INR in warfarin group and three doses combination groups were 1.40 and 2.49,3.34,4.46,compared with warfarin group and three combination groups,the difference was significant(all P <0.05).Compared with warfarin group and middle and high combination groups,the difference of concentration was significant(P <0.05,P <0.001).The PNS could enhance the AUC,AUMC,MRT,t1/2 and Cmax of warfarin in rats,but had no effect on t CLz/F and Vz/F.Conclusion PNS can enhance the efficacy of warfarin,prolong warfarin metabolism in the body time.
