Abstract:
Objective The LC-MS/MS method and 2D-LC/UV method were used to compared the consistency and correlation of olanzapine serum concentration.Methods Quality control samples and 52 serum samples of olanzapine were detected by LC-MS/MS and 2D-LC/UV method respectively.The consistency of results was compared by Bland-Altman method,and the correlation was analyzed by Pearson correlation analysis and Passing-Bablok regression.Results According to the Bland-Altman method,the percentage of the limit can be obtained:96.15% LoA =(-1.53,3.81).And 96.15% difference value was within the limit and the consistency of the two methods was good.According to the Pearson linear regression analysis,Passing-Bablok regression equation was OLZLC-MS/MS =1.02 × OLZ2D-LC/UV-1.18 and correlation coefficient was 0.999 (P < 0.01) with good correlation.Compared to the middle and high serum concentration,the relative deviation testes by two methods in low serum concentration(< 15 ng · mL-1) was larger.Conclusion The 2D-LC/ UV method and LC-MS/MS method have the good consistency and correlation in detecting olanzapine serum concentration,which can be used for conventional therapeutic drug monitoring(TDM).
