Abstract:
Objective To develop a high performance liquid chromatographic (HPLC) method for determination of hydroxychloroquine in human blood.Methods Metronidazole was used as the internal standard.Blood sample 300 μL were extracted with 900 μL of ethyl acetate,followed by drying with nitrogen and re-dissolving with mobile phase 60 μL.The separation was carried out on a Venusil ASB C18 (4.6 mm × 250 mm,5 μm) column at 35 ℃.The mobile phase consisted of acetonitrile -20 mmol · L-1 HCOONH4 buffer solution (15:85) (pH adjusted to 3.0 by HCOOH).The flow rate was 0.8 mL · min-1 and the injection volume was 20 μL.The detection wavelength was 254 nm.The specificity,limit of quantitation and standard curve,precision and accuracy,stability and recovery rate were investigated.Results The calibration curve of hydroxychloroquine was linear in the range of 100-3000 ng· mL-1,f=0.56×10-1C+0.39×10-1(r=0.998 2).The RSDs ofintra-day and inter-day precisions for hydroxychloroquine were within 5.5 %.The average extraction recovery in blood was (76.08 ± 6.21)% (n =5) for hydroxychloroquine.Conclusion The method was simple,sensitive,and accurate,and could be used for the measurement of hydroxychloroquine concentration in human blood.
