Effect of rosuvastatin on expression of matrix metalloproteinase-9 and tissue inhibitor of metalloproteinase-1 following focal cerebral ischemia-reperfusion

Abstract:
Objective To observe the effect of rosuvastatin on the expression of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in the brain tissue of cerebral ischemia-reperfusion rats and discuss its cerebral protective role.Methods Rats were randomly assigned into three groups:sham operation group(sham group),model group and test group.The rats in the test group were lavaged with 5 mg · kg-1 · d-1 rosuvastatin for 10 consecutive days before the model setting-up.The rats in the sham group and model group were fed normally.The model rats with middle cerebral artery occlusion were prepared by the modified Zea-Longa suture method.After vascular occlusion 2 h,remove the suture to form reperfusion.The rats in sham group received similar surgical treatment,but didn't insert the suture.The levels of MMP-9 and TIMP-1 were measured by RT-PCR,immunohistochemistry and Western blotting.Results The expression of MMP-9 mRNA in model group and test group were (0.45 ± 0.04) and (0.31-± 0.04) respectively,and the expression of TIMP-1 mRNA in model group and test group were (0.39 ± 0.05) and (0.52 ± 0.06) respectively,the difference were statistically significant (P < 0.05).The positive cell number and protein expression of MMP-9 and TIMP-1 in model group and test group were also statistically significant (P < 0.05).Conclusion Rosuvastatin can protect the brain injury through adjusting the expression of MMP-9 and TIMP-1.
