Effects of colistin methanesulphonate on immune function of macrophages in rat

Abstract:
Objective To investigate the effects of colistin methanesulphonate (CMS) on the immune function of rat peritoneal macrophages.Methods The cell viability of macrophages in rat peritoneal macrophages suspension (as negative control group) and those treated with 25,50,100,125,250,500 μg · mL-1 CMS for 24 h was measured by MTI colorimetry.The secretion level of tumor necrosis factor-α (TNF-oα)was quantified by enzyme-linked immunosorbent assay after macrophages were treated with 50,125 μg · mL-1 CMS for 3,6,12,24h.The phagocytosis of macrophages was calculated by microsphere-based immunofluorescence assay,after incubation with 50,125 μg · mL-1 CMS for 3,6,12 h.Results The cell viability were 118.46%,124.72%,122.54%,46.64%,13.06% when macrophages were exposed to CMS 25,50,100,250,500 μg · mL-1,the differences were statistically significant compared with negative control group 100.00% (P < 0.01);the cell viability of 125 μg · mL-1 CMS was 102.60% and the difference had no significant compared with negative control group (P > 0.05).After administration 3h,the secretion level of TNF-oα of negative control group and CMS 125 μg · mL-1 dose group were 32.51 pg · mL-1,56.09 pg · mL-1,the difference was statistically significant between them (P < 0.05).After incubation 6h,the percentage of phagocytosis of negative control group and CMS 50,125 μg · mL-1 dose groups were 70.93%,82.03%,80.80% and the differences were statistically significant compared to negative control group respectively (P < 0.01).Conclusion CMS could improve the immune function of macrophages and exhibit immunomodulatory by increasing viability of macrophages,promoting secretion of cytokines and enhancing phagocytosis of macrophages.
